Nice guide, but I'm talking about a formula to calculate the song difficulty level (from 0 to 6). I'm looking for something like: total of notes of an instrument ÷ total of seconds.

You might try actually playing the song, and basing it off that. Something like if you can't pass it, give it a six? I don't know.
Honestly, it's all speculation at this point. If you think it's hard, rate it as hard. Or you could do what Rock Band 3 did: Take all the songs, then arrange them from easiest to hardest. Make a setlist, then change the songs in the order that you made up. It's not legit at all, but it's a start.

Length (minutes and seconds), notes in succession (notes per second at the highest point), and how often the notes are played. Having even one part in a song that's too difficult... that alone can change a song from a solid two to a hard three, or even a soft four.
To give you any more information, I'd have to have and play the chart, or at the very least, know what song you have charted. But in any case, I really hope that the information that I gave you helps you out.
